中药解毒化瘀Ⅱ方对急性肝衰竭大鼠白细胞介素表达的影响

  • 目的    分析解毒化瘀Ⅱ方对急性肝衰竭大鼠白细胞介素(IL)表达的影响。方法    将200只无特定病原体级雄性Wistar大鼠完全随机分为中药组、阳性药组、造模阳性组、空白组,各50只;中药组、阳性药组、造模阳性组腹腔注射D-半乳糖胺及皮下注射内毒素制备大鼠暴发性肝衰竭模型;中药组造模前5 d灌胃给予解毒化瘀Ⅱ方,阳性药组造模前1 h给予盐酸维拉帕米灌胃,造模阳性组和空白组造模前1 h给予等量蒸馏水灌胃。采用免疫组织化学法检测肝细胞再生程度,采用定量聚合酶链反应法及蛋白质印迹法检测肝组织IL-2、IL-4、IL-10表达水平。结果    空白组大鼠肝脏细胞中增殖细胞核抗原的表达量较高,造模阳性组明显低于空白组(503±103比3 395±615),差异有统计学意义(P<0.05);而阳性药组、中药组大鼠中增殖细胞核抗原的表达量(2 172±364、1 577±288)高于造模阳性组,差异均有统计学意义(均P<0.05)。空白组设为1.00,造模阳性组IL-2 mRNA(21.06±1.03)和IL-10 mRNA(2.68±0.48)表达水平明显增高(P<0.05);阳性药组和中药组IL-2 mRNA(2.04±0.43、10.02±1.21)和IL-10 mRNA(1.42±0.14、1.94±0.38)的表达水平低于造模阳性组,差异均有统计学意义(均P<0.05)。造模阳性组IL-2和IL-10蛋白表达水平较空白组明显增高(0.71±0.12比0.22±0.07、0.55±0.15比0.30±0.04)(P<0.05);阳性药组和中药组IL-2蛋白(0.26±0.06、0.52±0.09)和IL-10蛋白(0.14±0.07、0.33±0.08)表达水平低于造模阳性组,差异均有统计学意义(均P<0.05)。结论    解毒化瘀Ⅱ方能有效降低大鼠肝组织IL-2及IL-10水平,切合肝衰竭的病因病机。

  • Effect of Chinese medicine Jiedu Huayu Ⅱ recipe on expression of interleukin in rats with acute liver failure

    【Abstract】Objective    To analyze the effect of Chinese medicine Jiedu Huayu Ⅱ recipe on the expression of interleukin(IL) in rats with acute liver failure. Methods    Totally 200 specific pathogen free male Wistar rats were randomly divided into Chinese herb group, drug control group, model control group and blank group, with 50 rats in each group. Rats in Chinese herb group, drug control group and model control group were made fulminant hepatic failure models by intraperitoneal injection of D-galactosamine and subcutaneous injection of endotoxin. Chinese herb group had intragastric administration of Jiedu Huayu Ⅱ recipe 5 d before modeling; drug control group had intragastric administration of verapamil hydrochloride 1 h before modeling; model control group and blank group had intragastric administration of distilled water 1 h before modeling. Liver cell regeneration was detected by immunohistochemical method; expressions of IL-2, IL-4, IL-10 in liver tissue were tested by quantitative polymerase chain reaction and western blotting. Results     Expression of proliferating cell nuclear antigen(PCNA) in liver cells of rats in model control group was significantly lower than that in blank group(503±103 vs 3 395±615)(P<0.05); expression of PCNA in drug control group and Chinese herb group(2 172±364, 1 577±288) was significantly higher than that in model control group(P<0.05). Expressions of IL-2 mRNA (21.06±1.03) and IL-10 mRNA (2.68±0.48) in model control group were significantly higher than those in blank group(1.00, 1.00)(P<0.05). Expressions of IL-2 mRNA (2.04±0.43, 10.02±1.21) and IL-10 mRNA (1.42±0.14, 1.94±0.38) in drug control group and Chinese herb group were significantly lower than those in model control group(P<0.05). Expressions of IL-2 and IL-10 in model control group were significantly higher than those in blank group(0.71±0.12 vs 0.22±0.07, 0.55±0.15 vs 0.30±0.04)(P<0.05). Expressions of IL-2(0.26±0.06, 0.52±0.09) and IL-10(0.14±0.07, 0.33±0.08) in drug control group and Chinese herb group were significantly lower than those in model control group(P<0.05). Conclusion    Jiedu Huayu Ⅱ recipe can effectively reduce levels of IL-2 and IL-10 in liver tissue of rats with acute liver failure.
