盐酸多奈哌齐对阿尔茨海默病患者认知功能及氧化应激水平的影响

  • 【摘要】目的    探讨盐酸多奈哌齐对阿尔茨海默病患者认知功能及氧化应激水平的影响。方法    选取2016年8月至2019年3月中国人民解放军联勤保障部队第九〇三医院收治的100例初次接受治疗的阿尔茨海默病患者。按照随机数字表法将患者分为对照组和观察组,各50例。对照组在基础治疗基础上口服吡拉西坦片,观察组在对照组基础上口服盐酸多奈哌齐片治疗。治疗6个月后评定2组临床疗效及认知功能和氧化应激水平变化。结果    治疗6个月后,观察组总有效率明显高于对照组[82.0%(41/50)比62.0%(31/50)],差异有统计学意义(P<0.05)。观察组简易精神状态检查量表评分高于对照组,阿尔茨海默病评估量表认知次量表评分低于对照组[(22.2±2.1)分比(19.3±3.2)分、(17±4)分比(20±4)分],差异均有统计学意义(均P<0.05)。治疗6个月后2组超氧化物歧化酶、丙二醛水平均较治疗前降低,且观察组低于对照组[(54±3)kU/L比(66±4)kU/L、(3.1±1.4)μmol/L比(4.6±1.3)μmol/L];谷胱甘肽过氧化物酶水平较治疗前升高,且观察组高于对照组[(93±6)U比(80±5)U],差异均有统计学意义(均P<0.05)。结论    盐酸多奈哌齐能有效改善阿尔茨海默病患者认知功能,提高临床疗效,作用机制可能是改善患者机体氧化应激水平,减少神经氧化损伤。


  • 【Abstract】Objective    To investigate the effect of donepezil hydrochloride on cognitive function and oxidative stress in patients with Alzheimer disease. Methods    A total of 100 patients with Alzheimer disease admitted to the 903rd Hospital of Joint Logistics Support Force of the Chinese People′s Liberation Army from August 2016 to March 2019 were randomly divided into control group and observation group, with 50 cases in each group. The control group was treated with piracetam. The observation group was treated with donepezil hydrochloride and piracetam. Clinical efficacy, cognitive function and oxidative stress indicators were analyzed after 6 months of treatment. Results    Total effective rate in the observation group was significantly higher than that in the control group[82.0%(41/50)vs 62.0%(31/50)](P<0.05). After treatment, Mini-Mental State Examination score in the observation group was higher and Alzheimer Disease Assessment Scale-Congnitive Section score was lower than those in the control group[(22.2±2.1) vs (19.3±3.2), (17±4) vs (20±4)](both P<0.05). Levels of superoxide dismutase and malondialdehyde significantly decreased after treatment in both groups; the levels in the observation group were lower than those in the control group[(54±3)kU/L vs (66±4)kU/L, (3.1±1.4)μmol/L vs (4.6±1.3)μmol/L](all P<0.05). Level of glutathione peroxidase significantly increased after treatment and it was higher in the observation group than that in the control group[(93±6)U vs (80±5)U](P<0.05). Conclusion    Donepezil hydrochloride can effectively improve the cognitive function through regulating oxidative stress and reducing neuro-oxidative damage and improve the clinical efficacy in patients with Alzheimer disease.
