  • 目的研究血清微小RNA(miRNA)特异性表达与儿童扩张型心肌病(DCM)低左心室射血分数(LVEF)的相关性。方法选取201311月至20163月首都医科大学附属北京安贞医院小儿心脏中心住院的DCM治疗1年后LVEF<55%的患儿15例(观察组),另选择同期年龄、性别匹配的DCM治疗1年后LVEF55%患儿15例(对照组)。应用基因测序技术筛选2组血清中差异表达miRNA,实时荧光定量聚合酶链反应法验证2组患儿血清中差异表达miRNA的相对表达水平,分析LVEFmiRNA表达水平的相关性。结果miRNA基因芯片试验检测结果显示,对照组与观察组比较有156miRNA表达上调(差异表达倍数>2P0.05),前8个差异表达的miRNA分别为miR-26a-5plet-7g-5pmiR-126-3pmiR-142-5plet-7f-5pmiR-27a-3pmiR-27b-3pmiR-143-3p。实时荧光定量聚合酶链反应验证实验结果表明,观察组血清let-7g-5p miR-126-3p相对表达水平高于对照组[(1.60±0.21)比(1.40±0.31)、(1.63±0.28)比(1.36±0.31)],差异均有统计学意义(P<0.05)。线性回归分析结果表明,治疗1年后30DCM患儿血清中let-7g-5pmiR-126-3p相对表达水平与LVEF呈负相关(r=-0.437-0.481,均P<0.05)。结论miRNADCM患儿治疗后LVEF降低具有相关性,调控miRNA有助于减少低LVEF的发生,改善患儿预后情况。

  • ObjectiveTo study the correlation between the specific expression of serum microRNA (miRNA) and the prognosis of children with dilated cardiomyopathy (DCM) and low left ventricular ejection fraction (LVEF). Methods From November 2013 to March 2016, 15 dilated cardiomyopathy patients with LVEF<55% after one-year treatment admitted to pediatric heart center of Beijing Anzhen Hospital, Capital Medical University were enrolled as the observation group (15 cases). At the same time, serum of age-matched controls with LVEF55% after treatment was collected  as control group(15 cases). The differentially expressed miRNAs in the two groups of serum were screened by gene sequencing technology. The relative expression levels of the differentially expressed miRNAs in the serum of the two groups were verified by real-time fluorescence quantitative polymerase chain reaction(RT-PCR). The correlation between LVEF and miRNA expression level was analyzed.Results  The results   of miRNA microarray showed that 156 miRNAs in the control group were up-regulated compared with observation group (differential expression multiple>2, P<0.05). Top eight miRNAs were miR-26a-5p, let-7g-5p, miR-126-3p, miR-142-5p, let-7f-5p, miR-27a-3p, miR-27b-3p, miR-143-3p, respectively. The results   of RT-PCR showed that the level of let-7g-5p and miR-126-3p in the observation group were  higher than that in the control group (1.60±0.21)  vs (1.40±0.31), (1.63±0.28) vs (1.36±0.31) (P<0.05). The results   of linear regression analysis showed that the relative expression levels of let-7g-5p and miR-126-3p were negatively correlated with LVEF (r=-0.437, -0.481, both P<0.05).Conclusion There is an association between miRNA and left ventricular ejection fraction in children with DCM after treatment.
