白藜芦醇对氧化应激环境中人脐静脉内皮细胞衰老和增殖及凋亡的影响

  • 目的 探讨在氧化应激环境中人脐静脉内皮细胞(HUVEC)衰老、增殖及凋亡受白藜芦醇的影响。方法 选择生长良好并处于对数生长期的HUVEC并分为3组,对照组细胞正常生长繁殖,过氧化氢组加入100 μmol/L过氧化氢,过氧化氢+白藜芦醇组加入100 μmol/L过氧化氢+50 μmol/L白藜芦醇。计数HUVEC细胞经过β-半乳糖苷酶染色之后的阳性细胞数,应用Modifit软件模拟检测不同周期细胞比例,应用流式细胞仪检测3组细胞凋亡情况。比较3组细胞衰老率,G1期、G2/M期和S期的细胞比例及细胞凋亡率。结果 过氧化氢组的细胞衰老率、细胞凋亡率均高于对照组[(76.8±5.3)%比(26.0±2.3%(9.4±1.0)%比(4.9±0.6%],过氧化氢+白藜芦醇组的细胞衰老率和细胞凋亡率[(36.9±3.6)%(5.7±0.7)%]均低于过氧化氢组,差异均有统计学意义(均P<0.05)。过氧化氢组G1期的细胞比例高于对照组,而G2/M期、S期的细胞比例均低于对照组;过氧化氢+白藜芦醇组G1期、G2/M期的细胞比例低于过氧化氢组,而S期的细胞比例高于过氧化氢组,差异均有统计学意义(均P<0.05)。结论 白藜芦醇可以促进氧化应激环境中HUVEC增殖,同时又可以抑制其衰老与凋亡,白藜芦醇可以保护氧化应激环境中的HUVEC

  • Objective To investigate the effect of resveratrol (RSV) on the senescence, proliferation and apoptosis of human umbilical vein endothelial cells (HUVECs) under oxidative stress. Methods HUVECs with good growth and logarithmic growth period were divided into three groups. Cells in control group grew and propagated normally. 100 μmol/L hydrogen peroxide was added to the hydrogen peroxide group; 100 μmol/L hydrogen peroxide+50 μmol/L resveratrol was added to the hydrogen peroxide+50 μmol/L resveratrol group. After β-galactosidase staining, the positive cells were counted and the cell proportion of different cycles were simulated by Modifit software; the cells apoptosis were compared among the three groups by flow cytometry. The cell senescence proportion and apoptosis rate in G1, G2/M and S phases were compared. Results The cell senescence rate and apoptosis rate of the hydrogen peroxide group were higher than those of the control group (76.8±5.3)% vs (26.0±2.3)%, (9.4±1.0)% vs (4.9±0.6)%; the cell senescence rate and apoptosis rate of the hydrogen peroxide+resveratrol group (36.9±3.6)%, (5.7±0.7)% were lower than those of the hydrogen peroxide group; the differences were statistically significant (all P<0.05). The proportion of cells in G1 phase of hydrogen peroxide group was higher than that in control group; the proportion of cells in G2/M phase and S phase were lower than those in control group; the proportion of cells in G1 phase and G2/M phase of hydrogen peroxide+resveratrol group was lower than that in hydrogen peroxide group; the proportion of cells in S phase was higher than that in hydrogen peroxide group (all P<0.05). Conclusion sResveratrol can promote the proliferation of HUVEC in oxidative stress environment; it can inhibit the senescence and apoptosis. Resveratrol can protect HUVEC in oxidative stress environment.
