  • 目的 探讨T细胞免疫球蛋白黏蛋白3(TIM3)和半乳糖凝集素9(Galectin-9)在子宫内膜癌中的表达及其与复发转移的关系。方法 选取山西省人民医院2019年2月至2021年10月经手术治疗的124例子宫内膜癌患者,另选择本院同期124例良性子宫内膜增生患者为对照组。采用RNAscope技术检测TIM3和Galectin-9的表达情况,系统分析二者表达与患者术后复发转移的相关性。结果 在子宫内膜癌组织中,TIM3和Galectin-9的阳性表达率分别为37.1%(46/124)、29.8%(37/124),而在良性子宫内膜增生组织中均未检测到TIM3和Galectin-9表达(0/124)。在术后发生复发或转移的子宫内膜癌患者中,TIM3与Galectin-9的阳性表达率均显著高于未出现复发转移的患者(均P<0.05)。子宫内膜癌的复发转移与淋巴结转移(P=0.010)和肌层浸润(P=0.040)有关。TIM3预测子宫内膜癌复发转移的曲线下面积(AUC)为0.872(95%置信区间:0.837~0.926),其敏感度和特异度分别为86.7%和79.5%;而Galectin-9的AUC值为0.830(95%置信区间:0.814~0.905),敏感度为90.3%,特异度为64.5%。单因素和多因素Cox回归分析显示,TIM3阳性表达、Galectin-9阳性表达和淋巴结转移均可作为提示子宫内膜癌患者复发转移的独立影响因素(均P<0.05)。结论 TIM3和Galectin-9在子宫内膜癌组织中特异性表达,且二者的高表达与术后复发转移风险显著相关。

  • Objective To investigate the expression of T-cell immunoglobulin mucin-3 (TIM3) and galectin-9 in endometrial carcinoma and their relationship with recurrence and metastasis. Methods A total of 124 patients with endometrial carcinoma who underwent surgery in Shanxi Provincial People′s Hospital from February 2019 to October 2021 were selected, and 124 patients with benign endometrial hyperplasia in the same hospital during the same period were selected as the control group. RNAscope technique was used to detect the expression of TIM3 and galectin-9, and the correlation between their expression and postoperative recurrence and metastasis was systematically analyzed. Results In endometrial carcinoma tissues, the positive expression rates of TIM3 and Galectin-9 were 37.1%(46/124) and 29.8%(37/124), respectively, while no expression of TIM3 and Galectin-9 was detected in benign endometrial hyperplasia tissues (0/124). In patients with endometrial carcinoma who developed postoperative recurrence or metastasis, the positive expression rates of TIM3 and Galectin-9 were significantly higher than those in patients without recurrence and metastasis (all P<0.05). Recurrence and metastasis of endometrial carcinoma were associated with lymph node metastasis (P=0.010) and myometrial invasion (P=0.040). The area under the curve (AUC) of TIM3 in predicting recurrence and metastasis of endometrial carcinoma was 0.872 (95% confidence interval: 0.837-0.926), with a sensitivity of 86.7% and a specificity of 79.5%; the AUC value of Galectin-9 was 0.830 (95% confidence interval: 0.814-0.905), with a sensitivity of 90.3% and a specificity of 64.5%. Univariate and multivariate Cox regression analysis showed that positive expression of TIM3, positive expression of Galectin-9 and lymph node metastasis could be used as independent influencing factors for predicting recurrence and metastasis in patients with endometrial carcinoma (all P<0.05). Conclusion TIM3 and Galectin-9 are specifically expressed in endometrial carcinoma tissues, and their high expression is significantly associated with the risk of postoperative recurrence and metastasis.
