经皮耳缘迷走神经刺激改善阿霉素诱导大鼠心电活动异常的研究

  • 目的 探究经皮耳缘迷走神经刺激(tVNS)对阿霉素诱导大鼠心电活动异常的影响。方法 18Sprague-Dawley雄性大鼠完全随机分为对照组(n=6)、阿霉素组(n=6)、阿霉素+tVNS组(n=6)。阿霉素组和阿霉素+tVNS组使用阿霉素(5 mg/kg)每周腹腔注射1次,注射3周以建立慢性心脏损伤模型,对照组腹腔注射等量0.9%氯化钠溶液。阿霉素+tVNS组每日使用刺激电极夹于大鼠双侧耳廓的耳屏内侧,以1 mA方波、1 ms脉宽、20 Hz刺激频率进行电刺激,每日刺激 30 min,持续刺激6周,对照组和阿霉素组仅夹置电极但不进行电刺激。6周后检测各组大鼠的心电图参数并进行统计分析,观察其变化。结果 与对照组相比,阿霉素组大鼠心电图的QRS时限、QT间期、RR间期明显延长[(22.5±1.0ms比(20.0±0.5ms、(32.28±0.18ms比(29.61±0.51ms、(175±27ms比(143±20ms],心率显著减慢[(349±44)次/min比(425±58)次/min],与阿霉素组相比,阿霉素+tVNS组大鼠心电图QRS时限、QT间期、RR间期[(20.6±0.5ms、(30.19±0.62ms、(143±10ms]明显缩短,心率明显增加[(422±30)次/min](均P0.05)。结论 tVNS能够稳定阿霉素诱导的大鼠心电活动异常,并可能降低恶性心律失常的发生风险。

  • Objective To explore the effect of transcutaneous vagus nerve stimulation(tVNS) on doxorubicin (DOX) induced abnormal cardiac electrical activity in rats. Methods Eighteen Sprague-Dawley male rats were randomly divided into control group (n=6), DOX group (n=6), DOX+tVNS group (n=6). DOX group and DOX+tVNS group were intraperitoneally injected DOX (5 mg/kg) once a week for 3 weeks to establish a chronic heart injury model. The control group was intraperitoneally injected with the same amount of 0.9% sodium chloride solution. In the DOX+tVNS group, stimulating electrodes were clamped on the inner side of the tragus of the rats bilateral auricles daily, and electrical stimulation was performed with 1 mA square wave, 1 ms pulse width, and 20 Hz stimulation frequency. The stimulation was 30 min once a day for 6 weeks. The control group and the DOX group only clamped electrodes but did not perform electrical stimulation. After 6 weeks, all rats electrocardiogram parameters were detected and statistically analyzed to observe their changes. Results The electrocardiogram showed that the QRS interval, QT interval, RR interval in DOX group were longer than those in control group[(22.5±1.0ms vs 20.0±0.5ms,32.28±0.18ms vs 29.61±0.51ms,175±27ms vs 143±20ms, and heart rate in DOX group was lower than that in control group[(349±44times/min vs 425±58times/min(all P0.05).The QRS interval, QT interval, RR interval in DOX+tVNS group were shorter than those in DOX group[(20.6±0.5ms,30.19±0.62ms,143±10ms, and the heart rate in DOX+tVNS group was significanly faster than that in DOX gruop [(422±30times/min(all P0.05). Conclusion tVNS can stabilize the abnormal cardiac electrical activity induced by DOX in rats, and may reduce the risk of malignant arrhythmias.
